The trend of amylase research focus is screening amylase produced by thermophilic bacteria that grow from hot springs. Indonesia has a diversity of bacteria that come from natural resources, such as in hot spring. The purpose of this study was to determine bacteria that showed the amylase activity from hot spring in Natar, Lampung Province. The methods used in this study include: (1) collecting sample from Natar hot spring, (2) isolation of a single colony grow on agar medium of Luria Bertani (LB), (3) screening of thermophilic amylase-producing bacteria growing on agar medium of Starch-Luria Bertani, and (4) determination of amylolytic activity by DNS reagent in soluble starch substrate. Hot water samples were collected at Natar hot spring at pH of 7.0 and temperature of 45 °C. Thermophilic bacteria have been isolated at growth temperatures of 45 °C with a sample dilution factor of 101 on agar medium of LB. Isolate bacteria producing amylase has been isolated from Natar hot spring based on the clear zones in bacteria which is grown on agar medium of Starch-Luria Bertani. The result showed the amylolytic activity of a bacterial crude in 1% soluble starch substrate is 240.7267 U/mL. The result showed that the amylase-producing thermophilic bacteria was isolated in the Natar hot spring, Lampung Province. © 2023 Author(s).
