Abstract

The mangrove forest is a one-of-a-kind natural ecosystem with significant ecological and economic value. These unique ecosystems are essential for maintaining biodiversity. Assessing and monitoring biodiversity required precise taxonomic identification. DNA barcoding is a valuable method for identifying organisms and studying biodiversity. This method involves using a short-standardized gene to identify a given species by comparing nucleotide sequences in the DNA fragment. We applied this molecular approach to identify and confirm specific fish species in a mangrove ecosystem. This study was conducted in several parts of the mangrove ecosystem along the coast of Lampung. The sampling point is divided into two areas; Here, we used some representative samples from the Eastern Coast of Lampung (Way Kambas National Park) and Lampung Bay (Tarahan, Sari Ringgung, and Dewi Mandapa beach). PCR was used to amplify the COI target region using the primers FishF1 and FishR1. The phylogenetic tree was constructed using the Neighbor Joining (NJ) method. The sequence-based K2P Neighbor Joining (NJ) trees normally consolidated species in conformity with their taxonomic classifications. The COI datasets could delimitate 10 families that consist of 10 species. Any of the studied species' barcode data has been submitted to GenBank. The barcoding approach successfully identified and determined fish at the species level. © 2023 Author(s).
